Risk Assessment of Perfluoroalkyl Compounds (PFCS) in Water and Sediment Samples of Hanjiang River

Abstract Contamination profiles of 11 kinds of perfluorinated compounds (PFCS) were analyzed by using ultra high performance liquid chromatography-triple quadrupole mass spectrometry in water and sediment samples of Hanjiang River in dry and wet seasons. The results showed that 11 kinds of PFCS were all detected. The total concentrations of PFCS in water and sediment were 0.3–23.04 ng/l, 0–55.1 ng/g in dry season and 0.16–19.68 ng/l, 0.99–85.07 ng/g in wet season. The maximum concentration of PFCS was detected in Wuhan where Hanjiang River feeds into Yangtze River. Meanwhile,wuhan had the highest concentration of perfluorooctanoic acid (PFOA) with 22.52 ng/l in dry season and 12.52 ng/l in wet season. Perfluoroheptanoic acid (PFHPA) and perfluorohexanoic acid (PFHXA) were the dominant PFCS in sediment and the highest total concentration of PFCS was detected in Taocha. There was little difference in the concentration composition of ∑PFCS in sediment in both seasons. The
risk quotient method was used for the ecological risk assessment of PFOA, perfluorooctane sulfonic acid (PFOS), perfluorononanoic acid (PFNA), PFHXA and perfluorodecanoic acid (PFDA) in water, and PFOA and PFOS in sediment based on the measured environmental concentration (MEC) and the predicted non-effect concentration (PNEC) of target pollutants.the assessment results suggest that the above PFCS in water and sediment of Hanjiang River posed no ecological risk to environment. 全氟化合物(perfluoroalkyl compounds, PFCS)是一类新型的持久性有机污染物, 最具代表性的物质是全氟辛烷(PFOA)和全氟辛烷磺酸盐(PFOS)。由于 PFCS 的碳氟键能大(466 kj/mol), 因此很难通过物理、化学、生物过程降解[1–2]。PFCS 一端连着烷基,具有疏水性, 一端连着羧基或磺酸基, 具有疏油性,具有很好的表面活性, 可作为表面活性剂。PFCS 广泛用于工商业及生活用品, 如消防泡沫、纺织、造
[3–4]纸、电镀、氟聚合材料等 。根据 PFCS 的连接的官能团不同, 分为离子型(以全氟羰基酸和全氟烷基磺酸为主)和非离子型(以氟调聚醇为主)化合物。离子型化合物主要存在于水和沉积物中(短链化合物倾向于水体, 长链化合物倾向于沉积物)[5–7], 非离子型化合物具有挥发性。另外, PFCS 具有生物富集性和生物毒性, 如肝毒性和神经毒性等[8–9]。对 PFCS 的迁移途径有两种假设: 一是海洋洋流的传播[10], 二是通过大气沉降的方式传播[11]。有研究报道, 在南北极的动物体内及大气中检测到 PFCS。
由于 PFCS 的广泛使用, 在地表水、地下水、沉积物、污水处理厂的进出水以及污泥、动物和人体的组织器官和血液中都检测到 PFCS , 浓度在 ng/l至 μg/l 级别[5–7,10–14]。
汉江是长江最大的支流, 流经 6 个省份, 经武汉汇入长江, 流域面积为 159 km2, 承载流域内上百万人口的用水需求, 其中上游的丹江口水库是南水北调中线工程的水源地, 是北京居民生活用水的重要地表水来源。汉江武汉段长达 62 km。武汉是全国重要的工业城市, 周边存在众多的氟加工企业及工业园区。目前关于汉江的研究不多, 且集中于对 PFOA 和 PFOS 两种典型 PFCS 的研究。本研究拟通过采集汉江旱季和雨季的水样和沉积物, 评估汉江流域 11 种 PFCS 的污染状况, 对该流域进行风险评价。
1实验部分1.1样品的采集及其前处理
汉江 7 个采样点(白河(BH)、陶岔(TC)、丹江
口(DJK)、襄阳(XY)、仙桃(XT)、集家嘴(JJZ)、武汉(WH))的位置如图 1 所示。样品采集于 2014 年旱季(3 月)和雨季(10 月)。水样储存于 5 L 聚乙烯瓶中, 加入浓硫酸调节 ph=3, 以抑制微生物活性,盖紧瓶盖(瓶盖内有聚丙烯垫片)。采集好的水样在0~4ºc 冷藏, 运输到实验室, 24 小时内处理完毕。沉积物采集表层 0~20 cm 的样品, 存储于 500 ml铝盒中, 运回实验室, −20ºc 冷藏。每个采样点的水样和沉积物各设置两个平行样。采样过程中所有设备、采样瓶和盖子需提前用甲醇、超纯水和河水润洗干净, 保证瓶内清洁, 不引入其他污染。在整个采样过程中防止接触聚四氟乙烯和玻璃制品。水样全部采用玻璃纤维滤膜过滤(GF/F 0.7 μm, Whatman, 英国)。取 2 L 滤液加入 50 ng 内标([13C8]-PFOS 和[13C8]-PFOA)。使用 Oasis HLB 固相萃取小柱(6 ml, 500 mg, Waters, 美国)进行固相萃取, 分别用 10 ml 甲醇和 10 ml 超纯水活化, 水样上样速度为 15 ml/min, 用 10 ml 超纯水淋洗,抽干 0.5 小时, 用 10 ml甲醇洗脱于聚丙烯管中, 用高纯氮气吹干, 用 1 ml 甲醇定容, 过 0.22 μm 的尼龙滤膜(安谱, 上海)后转移到聚丙烯的进样小瓶中。
沉积物样品冷冻干燥后, 采用超声辅助萃取[15]。取 2 g 土样, 加入 10 ng 上述内标, 用 10 ml 1%的醋酸涡旋振荡 10 秒, 放入 60ºc 超声波清洗机中振荡 20 分钟, 3000 rpm 离心 2 分钟, 将上清液转移到聚丙烯的烧杯中。在含沉积物的离心管中加入 5 ml 90%的甲醇, 涡旋 10 秒, 在 60ºc 的超声仪中振荡 20 分钟, 3000 rpm 离心 2 分钟, 转移到上面的离心管中。重复前面的操作一次, 最后再加入 10 ml 1%的醋酸润洗, 离心后的溶液再次转移到烧杯中,最后烧杯中的溶液体积为 40 ml。加入超纯水定容到 300 ml, 使得有机溶剂的含量低于 5%。后续处理同水样。
1.2 试剂与仪器
11 种 PFCS 的标品: 全氟己酸(Perfluorohexanoic acid, PFHXA)、全氟壬酸(Perfluorononanoic acid, Pfna)、全氟葵酸(perfluorodecanoic acid, PFDA)、
全氟丁烷磺酸(Perfluorobutane sulfonic acid, PFBS)、全氟己烷磺酸 (Perfluorohexane sulfonic acid, PFHXS)和全氟十一烷酸(Perfluoroundecanoic acid, PFUNDA)购自中国百灵威, 全氟庚酸(Perfluoroheptanoic acid, Pfhpa)和全氟十四烷酸(perfluorotetradecanoic acid, PFTEDA)购自英国 Fluorochem, 全氟辛酸(Perfluorooctanoic acid, PFOA)和全氟辛烷磺酸(Perfluorooctane sulfonic acid, PFOS)购自美国 Accustandard,全氟十二烷酸(Perfluorododecanoic acid, PFDODA)购自美国 Matrix, 两种同位素内标[13C8]PFOS和[13C8]-PFOA 购自英国 Cambridge Isotope Laboratories (CIL)。
仪器及分析条件: 超高效液相色谱三重四极杆质谱(UPLC-ESI-MS/MS, Agilent 1290-6460A, 美国)、ZORBAX Eclipse Plus C18 (Rapid Resolution HD 2.1×50mm×1.8-micron, Agilent, 美国)、超声波清洗机(SB25-12DT, 宁波新芝生物科技股份有限公司)、冷冻离心机(Hettich200r/320r, Universal, 德国)、固相萃取仪(Supelco, 美国)、氮气吹干仪(BF2000,八方世纪)。数据收集与处理使用 Masshunter和 Origin Pro 9.1 软件。液相系统所用色谱柱为 Agilent ZORBAX Eclipse
Plus-c18, 内径 1.8 μm, 2.1×50 mm, 前接 3 μm 在线 过滤器, 柱温为 40ºc, 进样量为 5 μl, 流动相为水(A)和乙腈(B), 流速为 0.25 ml/min, 初始比例为90% A和 10% B, 经 5分钟变为10% A和 90% B, 保持 1分钟, 6.1 分钟回到初始流动相保持 5 分钟。质谱条件: 电喷雾电离负源(ESI−), 雾化温度为 350ºc, 雾化器压力为 35 psi, 辅助气(N2)流量为 9 L/min, 毛细管电压为 3500 V, Delta EMV(−)为 400 V。多重反应检测模式(MRM)参数见表 1。
1.3 质量保证与质量控制
PFCS 的检测存在两个主要问题: 1) 采样及前处理过程中, 应避免与玻璃器皿(采样瓶、烧杯、容量瓶、进样瓶等)的接触; 2) PFOA 是聚四氟乙烯合成的助剂, 而很多 UPLC-MS/MS 的管线含聚四氟乙烯, 因此在检测时容易出现背景值的干扰。实验过程中设置空白针、溶剂空白及过程空白, 在进样过程中, 每分析 10 个样品进 1 次溶剂空白和过程空白。实验发现, 11 种 PFCS 的背景值皆小于仪器的检测限。按照 1.1 节的实验步骤, 分别以超纯水和沉积物为基质做过程空白及加标回收率。水样的加标浓度为 50 和 100 μg/l, 回收率为 70.2%~ 122%, 沉积物加标浓度为 25 和 50 μg/kg, 回收率为 40%~106.46%。在本研究中, 水样的方法检出限为 0.01~0.127 ng/l, 沉积物的方法检出限为 0.01~
0.127 μg/kg。采用内标法定量, 标准曲线的 R2 皆大于 0.999。
2结果与讨论2.1汉江水体中全氟化合物的浓度与组成
汉江水样中 11 种 PFCS 的浓度如图 2(a)所示。水样中∑PFCS 浓度为 0.16~23.04 ng/l。11 种 PFCS中, PFOA (含 8 个碳原子, 简写为 C8, 下同)和 PFBS (C4)的检出频率最高(71.43%~100%), 检出频率较低的为 PFHPA (C7), PFHXA (C6), PFDA (C10), PFOS (C8)和 PFUNDA (C11)(小于 28.57%)。汉江水样中,旱季 PFOA 的平均值为 3.9 NG/L(范围为<LOD~22.52 ng/l, LOD 为检出限), 雨季 PFOA 的平均值为 2.83 NG/L(范围为<LOD~16.52 ng/l), 所检测的样品浓度均低于饮用水的 PFOA 浓度限值(40 ng/l)[16]。旱季PFBS 的平均值为 0.162 ng/l (范围为 0.023~0.3 ng/l),雨季 PFBS 的平均值为 0.543 ng/l (范围为 0.035~ 1.85 ng/l)。旱季和雨季汉江水样中 11 种 PFCS 的浓度贡献略有不同, 表明不同季节可能存在不同的污染源。旱、雨季贡献率最大的为PFOA (0.3%~96%)和 PFDOA (0%~89%), 其次为 PFBS 和 PFDA, 而PFHPA, PFUNDA, PFOS 和 PFHPA 的贡献率较小。另外, 汉江上游以 PFDOA 为主要成分, 下游以PFOA 为主要成分。
从汉江上游至入长江口处, ∑PFCS 呈现升高的趋势, 且旱、雨季最高点皆出现在汉江注入长江的汇入口武汉, 可能是由于汉江接收蔡甸区污水处理厂(汉江最大的污水处理厂)排放的出水造成的。常规的污水处理工艺不能有效地去除 PFCS。Pan
[17]等 对北京 7 家污水处理厂 9 种 PFCS 的调查发现, 进水中 ∑PFCS 的浓度范围为 2.88~176 ng/l,出水中∑PFCS 的浓度范围为 5.48~498 ng/l, PFOA在进出水中的浓度最高。Sun 等[18–19]在沈阳和天津的污水处理厂都检测到 PFCS, 沈阳污水处理厂进、出水中∑PFCS 的浓度范围分别为 71.59~120.49 ng/l 和 44.52~60.07 ng/l, 而天津污水处理厂进、出水中 PFOA 的浓度范围分别为 20~170 ng/l 和30~145 ng/l。通常出水中的 PFCS 浓度高于进水[17], 主要原因是污水处理过程中, PFCS 从沉积物
[15]转移到水相 。因此, 污水处理厂出水是地表水中 PFCS 的可能污染源之一[18]。另外, PFBS 旱、雨季的贡献率为 4%~59%, 且检出频率为 100%, 最高浓度为 1.85 ng/l (武汉, 雨季), 说明汉江流域存在PFBS 的污染源, 同时也说明由于 2009 年斯德哥尔摩公约对 PFOS 的禁用和限制, 对 PFOS 的使用逐步过渡到 PFBS [20]。
在汉江没有检出 PFHPA, 而 PFDOA 的检出频率大于 43%, 与 Wang 等[21]的研究结果一致。值得注意
的是, 汉江流域的工业园区自从 2009 年开始迁移, 2012 年全部迁移完毕。本研究中∑PFCS的浓度低于其他文献中对汉江流域的检测结果[22], 但是与山东省南四湖、广东省珠江中 PFOA 的浓度相近[23–25]。
PFOS 的检出频率和浓度较低, 说明工业园区的迁移整顿以及 PFOS 的禁用降低了环境中 PFOS 的污染。这也解释了 2014 年汉江流域 PFCS 浓度较低的原因。金一和等[26]报道武汉地区地表水中 PFOA的浓度范围为 2.8~5.6 ng/l, PFOS 的浓度范围为
2.3~25.5 ng/l, 刘俊玲等[27]对武汉饮用水的调查中, PFOA的浓度范围是 17.25~119.31 ng/l, 高于本研究的检测结果, 而 PFOS 没有检出。
2.2 汉江沉积物中全氟化合物的浓度与组成
汉江沉积物中 11 种 PFCS 的浓度如图 2(b)所示, ∑PFCS 粗略地呈现正态分布, 最高点出现在陶岔, 这与 Wang 等[21]研究的汉江水体中 PFCS 的分布类似。沉积物中∑PFCS 的浓度为<LOD~85.07 ng/g, PFHPA 和 PFHXA 的检出频率最高, 均达到100%。旱季和雨季 PFCS 的检出频率略有不同, 旱季 11 种 PFCS 都有不同程度的检出, 而雨季 PFOA, PFNA, PFUNDA 和 PFDOA 的检出频率较低。汉江沉积物中, 旱季 PFHXA 的平均值为 14.23 ng/g (范围为 3.99~33.36 ng/g), 雨季 PFHXA 的平均值为9.29 ng/g (范围为 0.5~57.74 ng/g)。旱季 PFHPA 的平均值为 11.55 ng/g (范围为 4.02~19.02 ng/g), 雨季 PFHPA 的平均值为 4.63 ng/g (范围为 0.78~25.96 ng/g)。汉江沉积物中 11 种 PFCS 的组分贡献无明显季节差异(结果未列出)。沉积物中组分贡献率最大的是 PFHXA (37%~68%), 其次是 PFHPA (29%~ 44%), PFHXA 和 PFHPA 广泛存在于汉江的沉积物中, 这与汉江水样中以 PFOA 为主要成分不同。Guo 等[28]发现太湖沉积物中以 PFOS (9.98%~84.7%)和长链的全氟羰基酸(PFCAS)为主要成分。水体和沉积物中 PFCS 的浓度组成不同, 表明由于理化性质的差异, 不同的 PFCS 物质在水体和沉积物中的分配系数有差异。与其他文献相比, 本研究中沉积物中∑PFCS 的浓度高于太湖(1.11~8.21 ng/g)[28]、大
辽河(0.29~1.03 g/g)[29]和白洋淀(0.06~0.64 ng/g)[30], 与中国东部地区农村土壤中的数值范围(0.34~65.8
ng/g)相当。其他几种 PFCS 都有不同程度的检出, 但是对总浓度贡献较小, 长链的 PFCS (如 PFTA(C14))的浓度范围为 0.15~1.07 ng/g, PFTA 组分的贡献为0%~15%。
3 风险评估
水环境中的健康风险评价是通过计算风险熵值(risk quotient, RQ)进行评估, 是目前使用最普遍、最广泛的表征方法。RQ为目标物质检测到的浓度(measured environmental concentration, MEC)与预测无效应浓度(predicted non-effect concentration, PNEC)的比值: RQ=MEC/PNEC。
由于沉积物中缺乏 PFCS 的毒理学数据, 因此根据平衡分配法计算沉积物中预测无效应浓度(PNEC沉积物)。按照欧盟化学物质风险评价技术指导文件(TGD)[31]中标准环境特征参数, 忽略 PFOA 和PFOS 在水体、悬浮物与气体之间的分配, 参考曹莹等[32]得到 PFOA 和 PFOS 的 PNEC沉积物(mg/kg)。
水样中 PFOA, PFOS, PFNA, PFHXA 和 PFDA的 PNEC 值分别为 100, 25, 100, 97 和 11 μg/l, 沉积物中 PFOA 和 PFOS 的 PNEC 值分别为 2060 和 67 μg/kg[32–34]。风险评估分为 4 个等级: 较小风险(RQ<0.01)、低等风险(0.01<RQ<0.1)、中等风险(0.1<RQ<1)、高风险(RQ>1)。各采样点的 RQ 值列于表 2, 可以看出, 汉江水体中 5 种 PFCS 和沉积物中 PFOA 和 PFOS 的浓度尚未达到对生态环境造成危害的水平, RQ值远小于1。本研究结果与 Lin 等[35]使用 PNEC 对台湾南门河中 PFOA 和 PFOS 的风险
[36]评估结果以及 Gong 等 对沈阳大辽河流域PFOA 和 PFOS 的风险评估结果相同。但是, 沉积物中 PFOS 的 RQ 值相对较高, 考虑到 PFCS 具有生物富集性, PFOS 的生态风险仍需引起足够重视。由于 PFCS 的种类繁多, 其他 PFCS 在水体和沉积物中的生物学毒理数据仍很缺乏, 其生态风险尚需进一步研究。
4 结论
本研究采用超高效液相色谱–三重四级杆质谱仪检测 2014 年汉江水体和沉积物样品中 11 种PFCS 的浓度。结果表明, 水体中∑PFCS 的浓度范 围为 0.16~23.04 ng/l, 主要成分是 PFOA, 总浓度最高点出现在武汉。沉积物中∑PFCS 的浓度范围为<LOD~85.07 ng/g, 主要成分是 PFHPA 和 PFHXA,总浓度最高点出现在陶岔。采用熵值法对汉江PFCS 进行风险评估, 结果表明 PFCS 的浓度均未达到对生态环境造成危害的水平。伴随着 PFOS 和PFOA 的禁用以及汉江周边化工企业的迁移, 二者的污染有减轻的趋势。但是, PFBS 及其他替代品正在广泛使用, 缺乏其生产、使用和排放的相关信息, 对环境的长期潜在影响尚未可知, 因此针对替代品的污染状况、毒理效应及迁移转化规律, 应该开展更多的研究。
