Abstract：Based on such concepts as the theory of knowledge-based view，dynamic capability，and organizational learning field，and the supply chain data of 480 Chinese enterprises，the authors establish the theoretical frame and carry out the model analysis.Supply Chain Knowledge Management（SCKM）and Supply Chain Ambidexterity（SC Ambidexterity）is the important reflection of capability in supply chain management；and they promote innovation and operation performance of the enterprises. Accompanied with supply chain process，the capability of SCKM has significant positive impact on innovation and operation performance of the enterprise； after introducing supply chain exploration and supply chain exploitation（）into the relation between Supply Chain Knowledge Management Capability（SCKMC）and two different kinds of performance，the relation between SCKMC and SC Ambidexterity is significantly positive，and the role of explanation variables has been significantly improved；SC Ambidexterity partly plays the intermediary role in the relation between SCKMC and corporate operation performance，which means that the intermediary of SC Ambidexterity can strengthen the effect of corporate performance. Managers in the corporate should，first，pay more attention to acquiring，organizing，transforming，and exploiting，and work out the strategic plan of organizational information and knowledge；second，they should，in the cooperation with supply chain partners，do a better job in supply chain knowledge in advance，strengthen the innovative activities，and improve the efficiency of innovation；and third，they should encourage top managers to explore and analyze the potential and complex dynamic activities in both the up- and down- stream of the supply chain，change the paradigm of research on innovation，and expand innovation into all the departments of the enterprise.
