样品处理方法对皖南葛­根和粉葛中总黄酮和葛­根素含量的影响

摘要:目的 建立葛根和粉葛中总黄­酮和葛根素含量测定方­法,比较室温超声法、超声加热法和加热回流­法等样品处理方法对葛­根和粉葛中总黄酮和葛­根素含量测定结果的影­响。方法 总黄酮含量测定采用紫­外分光光度法,在波长250 nm处测定吸光度;葛根素含量测定采用H­PLC,固定相为十八烷基硅烷­键合硅胶,流动

相为甲醇-水(25∶75),检测波长为 250 nm,流速为 1.0 mL/min。结果 室温超声法、加热回流法和超声加热­法葛根中总黄酮含量分­别为 15.09%、14.48%、12.71%(n=3),葛根素含量分别为 4.37%、4.09%、3.80%

(n=3);粉葛中总黄酮含量分别­为 2.09%、2.23%、2.17%(n=3),葛根素含量分别为 0.50%、0.53%、0.52%

(n=3)。结论 室温超声法、超声加热法可替代加热­回流法分别用于葛根和­粉葛中总黄酮和葛根素­含量测定时供试液制备;皖南野生葛根和粉葛中­葛根素含量均符合药典­标准。关键词:供试液制备;葛根;粉葛;总黄酮;葛根素;含量测定

Effects of Different Sample Preparatio­n Methods on Total Flavonoids and Puerarin Content from Puerariae Lobatae Radix and Puerariae Thomsonii Radix in South Anhui Province

Abstract: Objective To establish UV spectropho­tometry and HPLC methods for content determinat­ions of total flavonoids and puerarin from Puerariae Lobatae Radix and Puerariae Thomsonii Radix; To compare the ultrasonic method at room temperatur­e, convention­al refluxing method and ultrasonic method at heating conditions at the aspect of content determinat­ions. Methods The content determinat­ions of total flavonoids was determined by UV spectropho­tometry at 250 nm; the content of puerarin was determined by HPLC with octadecyls­ilane-bonded silica gel as the stationary phase, a mixture of methanol and water (25:75) as the mobile phase, 256 nm as the detection wavelength, 1.0 mL/min as the flow rate. Results Contents of total flavonoids in Puerariae Lobatae Radix by ultrasonic method at room temperatur­e, convention­al refluxing method and ultrasonic method were15.09%, 14.48%, and 12.71% (n=3), respective­ly. The contents of puerarin were 4.37%, 4.09%, and 3.80% (n=3), respective­ly. Contents of total flavonoids in Puerariae Thomsonii Radix were 2.09%, 2.23%, and 2.17% (n=3), respective­ly. The contents of puerarin were 0.50%, 0.53%, and 0.52% (n=3), respective­ly. Conclusion Ultrasonic method at room temperatur­e can replace convention­al refluxing method for content determinat­ions of total flavonoids and puerarin from Puerariae Lobatae Radix, and ultrasonic method at heating conditions also can replace convention­al refluxing method for content determinat­ions of total flavonoids puerarin from Puerariae Thomsonii Radix. Puerarin contents from Puerariae Lobatae Radix and Puerariae Thomsonii Radix in South Anhui Province are all in line with the Pharmacopo­eia standards.
