参芪醒脑方对痴呆模型小鼠学习记忆能力及脑组织胆碱乙酰转移酶、乙酰胆碱酯酶含量的影响

摘要:目的 观察参芪醒脑方对氢溴酸东莨菪碱致痴呆模型小鼠学习记忆能力及脑组织胆碱乙酰转移酶(ChAT)、乙酰胆碱酯酶(AChE)含量的影响,探讨其防治老年痴呆的可能作用机制。方法 将 ICR 小鼠 110只随机分为对照组、对照+参芪醒脑方高剂量组、模型组、多奈哌齐组和参芪醒脑方低、中、高剂量组。对照组和模型组小鼠给予蒸馏水灌胃,其余各组小鼠给予相应药物灌胃,每日1次,连续 14 d。第 15 日 Morris 水迷宫实验、物体识别实验评价小鼠学习和记忆能力,行为学测试开始前20 min氢溴酸东莨菪碱腹腔注射诱发小鼠记忆障碍建立模型。Western blot检测小鼠皮层及海马组织ChAT、AChE 蛋白表达。结果 Morris 水迷宫

实验结果显示,与对照组比较,模型组小鼠逃避潜伏期明显延长(P<0.01);与模型组比较,参芪醒脑方中、

高剂量组小鼠逃避潜伏期明显缩短(P<0.05)。物体识别实验结果显示,与对照组比较,模型组小鼠对新事物

探究能力明显下降、分辨指数明显降低(P<0.001);与模型组比较,参芪醒脑方各剂量组小鼠分辨指数明显

升高(P<0.05,P<0.01,P<0.001)。Western blot结果显示,与对照组比较,模型组小鼠脑皮层及海马组织AChE蛋白表达明显升高(P<0.01);与模型组比较,参芪醒脑方低、中剂量组小鼠脑皮层AChE 蛋白表达明

显降低(P<0.01),参芪醒脑方各剂量组小鼠脑海马组织 AChE 蛋白表达明显降低(P<0.001),参芪醒脑方各剂量组小鼠脑皮层及海马组织ChAT蛋白表达均未见明显差异。结论 参芪醒脑方能明显改善氢溴酸东莨菪碱致痴呆模型小鼠的学习记忆能力,可能与降低脑内AChE蛋白表达有关。关键词:参芪醒脑方;阿尔茨海默病;东莨菪碱;胆碱乙酰转移酶;乙酰胆碱酯酶;小鼠
