Study on Quality Evaluation of Standard Decoction of Lonicerae Japonicae Caulis Decoction Pieces

Abstract: Objective To prepare standard decoction of Lonicerae Japonicae Caulis decoction pieces; To establish quality control method. Methods Fifteen batches of standard decoction of Lonicerae Japonicae Caulis decoction pieces were prepared according to the standardization method. With chlorogenic acid, caffeic acid and loganin as the detection indices, the extraction rate, content, and transfer rate were calculated and the UPLC characteristic chromatograms analysis method was established. Results According to the measurement of 15 batches of standard decoction of Lonicerae Japonicae Caulis decoction pieces, the extractum rate was at the range of 6.44%–11.95%. The contents of chlorogenic acid, caffeic acid and loganin were 12.17–22.61 mg/g, 2.75–5.12 mg/g and 21.71–40.32 mg/g, respectively. The transfer rates were 28.13%–52.25%, 53.93%–100.15% and 40.04%–74.36%, respectively. Besides, Similarity Evaluation System for Chromatographic Fingerprint of TCM (2012 edition) was used to analyze and compare the characteristic chromatograms. Ten common peaks were determined, and three chromatographic peaks of chlorogenic acid, caffeic acid, and loganin were identified. The similarity was over 0.96 by comparing with the control map. Conclusion This study establishes the quality control method of standard decoction of Lonicerae Japonicae Caulis decoction pieces, which is accurate, stable, and with good repeatability, which can provide references for the quality control of Lonicerae Japonicae Caulis formula granules.
