Study on Fingerprints of Flavonoids Ingredients in Epimedii Folium

Abstract: Objective To establish HPLC fingerprints of flavonoids ingredients in Epimedii Folium. Methods The chromatographic fingerprint was obtained with Agilent ZORBAX Eclispse SB-C18 (4.6 mm × 150 mm, 5 µm) as chromatographic column, with acetonitrile (A) - water (B) as the mobile phase for gradient elution (0–8 min, 15%→
25% A; 8–25 min, 25% A; 25–32 min, 25%→33% A; 32–48 min, 33%→38% A; 48–60 min, 38%→60% A; 60–70 min,
60%→85% A; 70~75 min, 15% A). The flow rate was 1.0 mL/min, and the column temperature was maintained at
30 ℃. The detection wavelength was set at 270 nm. The similarity analysis and methodological investigation were conducted to fingerprints of 11 batches of Epimedii Folium. Results There were 21 common peaks with similarities between 0.845 and 0.952 in 11 batches of Epimedii Folium and 5 chromatographic peaks were identified. The methodological investigation results showed that the established analytical method had good repeatability, precision and stability. Similarity analysis divided 11 batches of Epimedii Folium into 4 categories, consistent with the identification results of Epimedii Folium species. Conclusion Epimedii Folium from different producing areas have a certain of similarities. HPLC fingerprints can provide basis for species identification and intrinsic quality evaluation of Epimedii Folium.
