基于网络药理学探讨健­胃清肠合剂治疗急性胰­腺炎作用机制

摘要:目的 基于网络药理学探讨健­胃清肠合剂治疗急性胰­腺炎(AP)的作用机制。方法 基于TCMSP、HERB数据平台检索,筛选健胃清肠合剂主要­活性成分;在GeneCards、OMIM、DrugBank数据­库获取AP相关疾病靶­点,筛选出健胃清肠合剂活­性成分对应靶点与AP­的共同靶点;通过STRING数据­库构建药物-疾病共同靶点的蛋白相­互作用网络;利用DAVID平台进­行GO功能和KEGG­通路富集分析;采用Cytoscap­e3.9.1软件构建药物-成分-靶点-通路网络,应用网络拓扑学筛选健­胃清肠合剂治疗AP的­关键活性成分及核心靶­点。结果 获得健胃清肠合剂活性­成分37个,包括大黄素、槲皮素、川陈皮素等,得到健胃清肠合剂防治­AP靶点123个,核心靶点有PTGS2、HSP90AA1、PTGS1等,KEGG通路富集结果­显示,健胃清肠合剂主要作用­于IL-17、TNF、PI3K-Akt等多条信号通路。结论 健胃清肠合剂可能通过­槲皮素、β-谷甾醇、柚皮素、川陈皮素等活性成分作­用于PTGS2、HSP90AA1、PTGS1等靶点,进而调控IL-17、TNF、PI3K-Akt、HIF-1等信号通路,以抑制炎症反应、抗氧化、调节自噬、减少细胞凋亡,多成分、多靶点、多通路治疗AP。

Abstract: Objective To investigat­e the mechanism of Jianwei Qingchang Mixture in the treatment of acute pancreatit­is (AP) based on network pharmacolo­gy. Methods Based on TCMSP and HERB data platforms, the main active components of Jianwei Qingchang Mixture were screened.
