The Metropolitan Police acted after receiving a complaint within the last few days.
It relates to an incident which is alleged to have taken place in 1997.
Worboys, 60, has not been arrested or questioned but the new investigation could delay his release from prison.
If new charges are brought Worboys is likely to remain in custody.
The development raises hopes for victims who have been battling to prevent Worboys from being freed from jail.
Last night Scotland Yard said: “The Metropolitan Police Service is investigating an allegation of nonrecent sexual assault which was reported to police in January 2018.

Worboys, of Rotherhithe, southeast London, was jailed indefinitely in 2009, with a minimum term of eight years, for attacks on 12 women involving rape, assaults and drugging. He is suspected of attacking more than 100 others.
But the Parole Board sparked fury when it decided to free the serial sex attacker just nine years after he was jailed and without informing the women he had attacked. There was fresh anger last night as it emerged that ahead of Worboys’ release from prison he may have been secretly moved from high-security HMP Wakefield in West Yorkshire to the capital – where the vast majority of his victims were attacked.
It is believed Worboys was driven the 200 miles to Belmarsh prison in south-east London at the weekend.
Lawyers for victims described the development as “extremely distressing” for them.
Richard Scorer – a lawyer at Slater and Gordon which represented a number of Worboys’ victims – said: “It’s adisgrace that victims are perpetually ignored by the authorities and are not consulted about his release.”
Tory MP Zac Goldsmith said the proposed release of Worboys has “absolutely horrified and terrified many, many victims”.
He added: “They are appalled to learn today that he has been moved to London’s Belmarsh prison.”
A group of 27 MPs has called for an “exclusion zone” to be imposed to ban Worboys from the capital.
