调心方对 APP/PS1 双转基因AD小鼠空间­学习记忆能力和海马突­触传递长时程增强的影­响

摘要:目的 观察调心方对 APP/PS1 双转基因阿尔茨海默病(AD)小鼠空间学习记忆能力­和海马突触传递长时程­增强(LTP)的影响,探讨其作用机制。方法 将 54 只 3 月龄 APP/PS1 双转基因AD小鼠随机­分为模型组、调心方组和阳性药组,以同月龄同性别 18 只 C57BL/6J 野生小鼠作为正常组,各给药组给予相应药物­灌胃。给药12周后,进行 Morris水迷宫行­为学检测和离体电生理­记录。结果 Morris 水迷宫测试结果显

示,定位航行实验与模型组­比较,调心方组逃避潜伏期时­间明显缩短(P<0.01);空间探索实验与模型组­比较,

调心方组小鼠穿越原平­台所在象限次数和在原­平台所在象限停留时间­明显增加(P<0.05,P<0.01)。在离体

海马电生理诱导中,与模型组比较,调心方组海马兴奋性突­触后电位斜率明显增强(P<0.01),海马双脉冲易

化无明显差异(P>0.05)。结论 调心方可有效改善 APP/PS1 双转基因 AD 小鼠空间学习记忆能力­和神经突触可塑性,具有认知保护作用。

Effects of Tiaoxin Formula on Spatial Learning and Memory Ability and LTP in Hippocampu­s of APP/PS1 Double Transgenic Mice with Alzheimer Disease

Abstract: Objective To investigat­e the effects of Tiaoxin Formula on spatial learning and memory ability and long-term potentatio­n (LTP) in the hippocampu­s of APP/PS1 transgenic mice with Alzheimer disease; To discuss its mechanism of action. Methods Totally 54 three-month-old APP/PS1 transgenic mice were randomly divided into model control group, Tiaoxin Formula group and positive control group, with 18 mice in each group. Another 18 three-month-old C57BL/6J wild mice were chosen as normal control group. All administra­tion groups receive relevant medicine. 12 weeks later, Morris water maze test was used to test behavior and in vitro electrophy­siology record. Results The Morris water maze test showed that in place navigation test, compared with the model control group, escape latency time in Tiaoxin Formula group was significan­tly reduced (P<0.01). In the space experiment­s, compared with the model control group, the number of crossing the platform quadrant and the time staying at the platform quadrant in Tiaoxin Formula group were significan­tly increased (P<0.05, P<0.01). In vitro hippocampa­l electrophy­siological induction, compared with the model control group, fEPSP slope in the hippocampu­s in Tiaoxin Formula group were significan­tly increased (P<0.01), the PPF had no significan­t difference (P>0.05). Conclusion Tiaoxin Formula can improve the spatial learning and memory ability and LTP of APP/PS1 transgenic mice with Alzheimer disease, thus can realize cognitive protection effects.
