HPLC指纹图谱结合­主成分分析对不同产地­头顶一颗珠质量的评价

1.首都医科大学中医药学­院,北京 100069;2.中医络病研究北京市重­点实验室,北京 100069摘要:目的 通过 HPLC指纹图谱结合­主成分分析(PCA)对不同产地头顶一颗珠­药材的质量进行评价,为其质量控制提供依据。方法 采用 Hibar C18 色谱柱(4.6 mm×250 mm,5 µm),乙腈-水为流动相进行梯度洗­脱,流速为 1.0 mL/min,检测波长为 203 nm,柱温为 30 ℃,利用相似度评价软件和­PCA 对 14批头顶一颗珠药材­检测结果进行分析比较。结果 HPLC指纹图谱的方­法学考察符合相关要求,共标定了头顶一颗珠药­材HPLC指纹图谱的­15 个共有峰,14批药材的相似度为 0.389~0.979。PCA 结果显示前3个主成分­的累积方差贡献率为 87.674%,S2 样品综合得分最高(4.926)、质量最好。结论 所建立的 HPLC 指纹图谱结合 PCA 可以客观、有效地评价不同产地头­顶一颗珠药材的质量。关键词:头顶一颗珠;指纹图谱;高效液相色谱法;主成分分析;相似度分析

Evaluation of Quality Coherence of Trillium tschonoski­i Maxim. from Different Producing Areas Based on HPLC Fingerprin­t and PCA

Abstract: Objective To evaluate the quality coherence of Trillium tschonoski­i Maxim. from different producing areas by HPLC fingerprin­t and PCA; To provide a method for quality control. Methods Samples were separated by Hibar C18 (4.6 mm × 250 mm, 5 µm) with acetonitri­le-water as gradient mobile phase at the flow rate of 1.0 mL/min. The wavelength was 203 nm and the temperatur­e was 30 ℃. Chromatogr­aphic Fingerprin­t Similarity Evaluation System and PCA were used to analyze the data. Results The results of method validation of HPLC fingerprin­t met technical standards. 15 common peaks was verified and the similariti­es of 14 batches of Trillium tschonoski­i Maxim. from different producing areas were among 0.389–0.979. 3 principal components with the characteri­stic root cumulative contributi­on rate reaching 87.674% were screened out by PCA results. The composite score of S2 was the highest (4.926), and the quality was the best. Conclusion The applicatio­n of HPLC combined with PCA can objectivel­y and effectivel­y evaluate the quality difference of Trillium tschonoski­i Maxim. from different producing areas.
